Os Chefões traz a saga da família Tempio, numa trama sangrenta que leva às últimas consequências uma história de traição e ódio entre famílias mafiosas ítalo-americanas da década de 30. O frio e calculista Ray (Christopher Walken) lidera a família de três irmãos. Johnny (Vincent Gallo), o caçula do grupo, sempre tem suas atitudes remediadas pelo irmão mais velho. Chez (Chris Penn), o irmão do meio, já dá sinais de loucura por seu passado criminoso. Para eles, construir fortuna em cima de cadáveres sempre foi comum, até virem no caixão o corpo do jovem Johnny. Os Tempio já nutriam uma rixa antiga contra Gaspar Spoglia (Benicio Del Toro), que se agrava quando no funeral de Johnny recebem uma coroa de flores do arqui-inimigo. Seria esse um sinal, que resultaria no início de uma série de crimes por vingança?
