Sean Jones é um adolescente que presenciou uma brutal execução ligada ao crime organizado, sendo a testemunha-chave daquele que a mídia chama como sendo o julgamento do século. Ele é encaminhado ao tribunal pelo agente do FBI Nelville Flynn, que o leva de avião até Los Angeles. Entretanto o que eles não sabem é que a máfia contratou Yakama, um assassino profissional, para impedir que Sean chegue ao seu destino. Para atingir seu objetivo ele decide liberar centenas de serpentes venenosas dentro do avião, o que logo faz com que tripulação e passageiros entrem em pânico.
