O filme recria a Expedição Científica Roosevelt-Rondon (1913-1914), comandada pelo coronel brasileiro Cândido Mariano da Silva Rondon e o ex-presidente americano Theodore Roosevelt. Juntos, eles atravessaram o Pantanal e a floresta amazônica para explorar o misterioso rio da Dúvida. Valendo-se de dramatização, material de arquivo original da Comissão Rondon e do acervo Memória Civelli, além de depoimentos das netas de Rondon e de caciques de diversas etnias, os dois personagens principais da história “refazem”, após um século, a expedição, apresentando suas diferentes visões de mundo e refletindo sobre meio ambiente, política indigenista e desenvolvimento da Amazônia.
