Peter Bretter (Jason Segel) é um compositor que está arrasado desde que Sarah Marshall (Kristen Bell), sua namorada de cinco anos, terminou o relacionamento. Para levantar o ânimo, seu meio-irmão Brian (Bill Hader) sugere uma viagem ao Havaí. Peter aceita a ideia e parte para um resort na ilha de Oahu, só que logo ao preencher o cadastro no hotel vê Sarah com seu novo namorado, o roqueiro inglês Aldous Snow (Russell Brand). Peter mais uma vez cai na fossa, sendo salvo por Rachel (Mila Kunis), uma atendente do hotel que o convida para um luau.
